Please confirm if you are able to optimize other drives on your PC.

If so, I suggest you to reinstall AVG TuneUp program and check if you experience the same.

I'm trying to optimize my C drive with TuneUp and it keeps telling me I don't have enough disk space. I have 34.5gb free - how much space does it need?? I had no issue on my other computer so I don't understand what the problem is.
